Pyruvate Sensor v2 w/ Gemini

Similar to BBa_M36701, this is a coupling of the Pyruvate sensor (BBa_M36698) with the Gemini Protein Generator (BBa_M36709)

IclR binds with pyruvate to form a more stable structure to bind to the inhibitory regulation sites on the aceBAK promoter. By combining the IclR gene and the aceBAK promoter into the same plasmid, the presence of IclR and pyruvate will inhibit the aceBAK promoter and any genes downstream of it, in this case Gemini. While there will be some residual transcription in the presence of only IclR, the presence of pyruvate increases the stability of binding with the aceBAK promoter region so inhibitory activities are higher. We choose to order to pieces of DNA to test the influence of signal leveling between the frequency of the IclR gene and aceBAK promoter. In this composite, the IclR gene is occurring in an equal frequency as the aceBAK promoter. See the Part Design for more information accounting for the ability of IclR to inhibit the aceBAK promoter.
